EFFECTS OF ~IV VITRO INCUBATION OF BOVINE SPERMATOZOA IN BOVINE FOLLICULAR FLUID l
نویسنده
چکیده
Addit ion of either 25% or 50% bovine follicular fluid (FF) to Krebs Ringer phosphate buffer, with .5% fructose added (KRPF), maintained higher percentages (P<.05) of motile sperm throughout 9-hr incubation at 37 C than did 100% KRPF or 100% FF. Accelerated sperm moti l i ty and head-to-head agglutination of sperm cells were consistently observed in all diluents that contained FF. The percentage of morphologically normal cells was not affected by any combination of FF and KRPF or by incubation. All concentrations of follicular fluid accelerated alteration of the acrosome with most of the alteration being apparent by 3 hr of incubation. Sperm treated with follicular fluid exhibi ted evident disintegration or detachment of the acrosome while cells t reated with KRPF exhibited a slight thickening and roughening of the acrosome. (

Highly Efficient In Vitro Production of Bovine Blastocyst in Cell-Free Sequential Synthetic Oviductal Fluid vs. TCM199 Vero Cell Co-Culture System
Background The aim of this study was to establish a cell-free sequential culture system that can support high levels of in vitro embryo development and blastocyst formation from bovine zygotes. To this end, this investigation was carried out to evaluate the effects of glucose, serum and EDTA on bovine zygote in vitro development. MaterialsAndMethods Bovine presumptive zygotes were derived from ...
متن کاملEffect of Goat Follicular Fluid on in vitro Production of Embryos in Black Bengal Goats
The study was undertaken to elucidatethe beneficial effects of goat follicular fluid (gFF) added to maturation and culture media on in vitro maturation, fertilization and post-fertilization development of oocytes in Black Bengal goats. Follicular fluid and oocytes were collected from slaughter house goat’s ovaries using the aspiration technique. Media were prepared using TCM-199 supplemented wi...
متن کاملTransfection of bovine spermatogonial stem cells in vitro
Spermatogonial stem cells (SSCs) are the only stem cells in adults that can transfer genetic information to the future generations. Considering the fact that a single SSC gives rise to a vast number of spermatozoa, genetic manipulation of these cells is a potential novel technology with feasible application to various animal species. The aim of this study was to evaluate enhanced green fluoresc...
متن کاملO-13: Gene Expression Pattern of Adiponectin and Adiponectin Receptors in Bovine Dominant and Non-Dominant Follicles
Background: Recent studies have revealed that adiponectin, a novel adipocytokine, and its two receptors (AdipoR1 and AdipoR2) are involved in regulation of ovarian function in various farm animal species. The aim of this study was to investigate the gene expression pattern of Adiponectin and its receptors in ovarian cells of dominant (DFs) and non- dominant follicles (NDFs) in the bovine. Mater...
متن کاملA comparison study of insulin concentrations in follicular fluid, serum and in vitro-production of bovine embryos – risks of generating unfavourable metabolic conditions during early development
Methods and results Eleven in vivo-studies measuring insulin in serum or follicular fluid have been compared to nine IVP-protocols using insulin. In vitro concentrations were much higher than in vivo while not much is known about the different activity and stability of insulin in vitro versus in vivo. We measured insulin concentrations before and after maturation in an experimental trial using ...
